What was done
Samples of poison were collected from live Incilius alvarius (Colorado River toads) in the Sonoran Desert, Arizona, USA. Tryptamine derivatives were extracted using acetone/water (70/30, v/v). To enable detection of trace components without detector overload, 5-MeO-DMT was chromatographically separated prior to qualitative evaluation. Samples were analyzed using attenuated total reflection-Fourier transform infrared spectroscopy (ATR-FTIR), gas chromatography-mass spectrometry (GC-MS), high-performance liquid chromatography-quadrupole time-of-flight high-resolution mass spectrometry (HPLC-qToF-HRMS), and HPLC-tandem mass spectrometry (HPLC-MS-MS). The quantitative method was validated.
What was found
ATR-FTIR and MS methods confirmed 5-MeO-DMT as the major constituent (mean ± SD: 410,000 ± 30,000 µg/g). Other identified tryptamines included bufotenin (2,800 ± 1,900 µg/g), 5-MeO-tryptamine (490 ± 260 µg/g), 5-HO-N-methyltryptamine (270 ± 120 µg/g), and DMT (250 ± 80 µg/g), with high variability between individual samples. Additional confirmed compounds were 5-MeO-N-methyltryptamine, 5-MeO-tryptophol, 2-(5-methoxy-1H-indol-3-yl)-acetic acid (5-MIAA), and tryptophan. 5-MeO-tryptamine and two positional isomers of hydroxylated MeO-DMT were detected in I. alvarius poison for the first time.
Why it matters
This study provides validated qualitative and quantitative reference data on the chemical profile of Incilius alvarius poison, assisting forensic identification and toxicological characterization of toad venom products.
Limits
The total number of toad specimens sampled was not reported in the abstract. Samples were obtained from a single geographic region (Sonoran Desert, Arizona), precluding assessment of geographic or seasonal variation in alkaloid composition.
